    Antibody services and custom productions

  1. What is the delivery schedule for a contract production?
    The delivery of the purified antibodies from a standard CL1000 flask production run is 8-10 weeks after signing of the contract. This entails 1-2 weeks of expansion of cells and establishment of the CL1000 flask, 5 weeks of production and harvesting and 1-2 weeks down-stream processing and QC. If a larger amount is required, the CL1000 production can be continued for an additional 1-5 weeks or several flasks can be run in parallel.
    For large-scale productions bimonthly deliveries will be the best option, giving first delivery 8-10 weeks after the production run is started.

  2. How much antibody can I expect from one standard CL1000 production?
    The yield depends on the productivity of the clone. The concentration of antibodies in the harvest supernatant is approximately 50 times the concentration in a cell culture supernatant, and a standard production run gives approximately 100-120mL harvest supernatant. The antibody is purified using a suitable purification resin according to isotype and origin of the antibody, to optimise the final yield. Read more.

  3. My hybridoma is growing in a serum free medium, do you do productions using serum free medium?
    Diatec Monoclonals standard medium is a specially adapted RPMI1640 with additional L-Glutamine and glucose, added foetal bovine serum of FDA approved origin. However, we have had several successful productions using different serum free media on customers’ request.

  4. I have an antigen against which I would like to establish an antibody. Can Diatec Monoclonals help me?
    Diatec Monoclonals has several years of experience in establishing hybridomas from customers’ antigens. Our hybridoma production projects are tailored to suit each customer (e.g. mouse strain, number of immunisations etc.) and our programs are modular so the customer can choose how extensive each phase of the program should be.   5. I have a hybridoma from which I would like to produce monoclonal antibodies, but I do not know anything about it regarding productivity, subclass etc. Can Diatec Monoclonals help?
    Diatec Monoclonals AS has well established systems for analysis for the most crucial parameters that ensure optimal production. On request we can perform analysis of antibody productivity of cell culture supernatant, determine the percentage of producing cells, subclass and specificity analysis and other assays. Full certificates of analysis are supplied.

  6. I need a small amount of antibody now for development purposes, with a potential scaling-up at a later date.
    Diatec Monoclonals in vitro production systems are adaptable to suit different production scales. The Integra Celline CL1000 production scale will cover your initial research needs. The use of our in vitro production technology will ensure that the products delivered from this production scale will be of the same high, stable quality as products in larger scale systems (Maximizer) when you are ready to scale up.   7. Do you perform custom conjugations of antibodies to fluorochromes and enzymes?
    Different conjugation methods are one of Diatec Monoclonals special fields of expertise. We have extensive experience in conjugating antibodies to fluorochromes and enzymes, among them FITC/fluorescein, phycoerythrin (PE), allophycocyanin (APC), biotin, horse radish peroxidase (HRP) and alkaline phosphatase (ALP).

  1. Why are all productions at Diatec Monoclonals performed in vitro?
    After Köhler and Milstein's discovery of hybridomas producing monoclonal antibodies in 1975, production was initially done in the ascitic fluid of production animals. Hollow fibre technology was developed over the same period, and has over the last decade given us an excellent alternative to using production animals. Due to animal welfare concerns, and the possibility of obtaining a pure product, without animal contaminants, with high batch to batch reproducibility, hollow fibre technology has proved to be a very reasonable choice.   3. What are the advantages of using monoclonal human IgE compared to IgE from a donor patient?
    Diatec Monoclonals Human IgE is unique in that it is produced in vitro from a hybridoma. This unique source guarantees freedom from contamination by other immunoglobulin isotypes. The original IgE producing cell was obtained from a healthy donor tested negative by US-FDA approved tests for HIV, HCV and Hepatitis B. The purity of the product is >98%, tested by SDS-PAGE.

    This antibody is well suited as a standard in IgE quantifying assays due to its very low batch-to-batch v ariation. Other applications include immunochemistry and cellular immunology research. In contrast to IgE from a myeloma patient, where batch-to-batch variability may be high, the monoclonal source of Diatec IgE ensures very high reproducibility.

  4. Does the product no. 5010, anti-human C5b-9 antibody bind both the soluble and membrane bound form of the TCC complex?
    This antibody recognises C5b-9, the terminal complement complex (TCC) and is directed against a neoepitope exposed in C9 when incorporated into the terminal complement complex (TCC). The clone aE11 produces monoclonal antibodies against neoantigens of both membrane bound (MAC) and fluid phase (SC5b-9) complexes. Read more.

  5. Is it possible to order bulk amounts of your list products?
    Should you require a larger amount (from 10 to several hundred milligrams) of our unconjugated products, please contact us for a quote. We can offer substantial discounts to our list prices on bulk purchases, and can adapt the concentration, buffer etc. to suit your application. Read more.

  6. I would like to test an antibody in my own system. Can I have a test sample of your antibodies?
    All of our conjugated products are available in 10-test aliquots for you to test for performance in and compatibility with your assays and systems.

  7. What is the difference between the two anti-human CD3 clones you have?
    Diatec Monoclonals AS stocks two different clones against the leukocyte marker CD3, UCHT-1 and CRIS-7. The antibody from clone UCHT-1 is your best choice for applications such as frozen tissue sections, histology, RIA, immunoprecipitation and flow cytometry, whereas the CRIS-7 product is well suited for purposes such as stimulation of T-cells. It is observed that the epitope for CRIS-7 antibody may be blocked or denatured by fixation.

  8. Should I order with or without azide?
    Sodium azide (NaN3) is added to our products as a preservative at a concentration of 0.09%. If your application involves live cells or enzymes sensitive to sodium azide (e.g. horse radish peroxidase) you should order an azide free product. Azide free products are denoted by ?a at the end of the product number.

  9. How are my products shipped?
    Products from Diatec Monoclonals AS are shipped by FedEx. This ensures efficient, door-to-door delivery of your order.
    Should you wish to have your product shipped by a different courier company, we will be happy to arrange this, but reserve the right to ship EXW Oslo. See list of delivery costs.
